Poster #124 Species differentiation of Dictyocaulus viviparous and Dictyocaulus eckerti

Bovine lungworms (Dictyocaulus viviparous) were isolated cattle in Missouri, Wisconsin, Mississippi and New Zealand. Also lungworms were isolated from captive red deer (Dictyocaulus eckerti) and white-tailed deer (Dictyocaulus capreolus). Differentiation would be carried out by way of polymerase chain reaction (PCR) by amplifying the ribosomal second internal transcribed spacer (ITS2). The key of this research was better to understand and determine materials and methods that would allow us to effectively amplify the ITS2 gene of each species for analysis. Several GC amplifiers were discovered (ethylene glycol and 1,2-propanediol) as well as promega T-vector as a cloning kit which would allow us to make numerous copy of the gene of interest. |
